Scientists have discovered a new mechanism that causes hair follicle stem cells to lose their ability to regenerate.
There are many mysteries surrounding the biological processes that lead to hair loss in humans, but scientists continue to make discoveries that unravel these mysteries and establish technologies that may one day cure them. The study, conducted by scientists at Tokyo University of Medicine and Dentistry and Tokyo University, looked at how hair follicle stem cells help new hair grow and why the relationship between them is broken.
The stability of hair follicle stem cells depends on a healthy combination of two types of cell division. In periodic symmetrical cell division, hair follicle stem cells divide into two types of cells with the same fate, while in asymmetric cell division, they divide into a distinct cell with a separate, self-renewing stem cell.
“Symmetrical and asymmetric cell divisions must be balanced for proper tissue function,” says study author Amy Nishimura. When stem cells preferably experience one of these, or worse, detach from the normal process of cell division, the limbs suffer pain. “In this study, we want to find out how stem cell division helps hair grow as it ages.”
The scientists performed experiments with young and old mice, in which they monitored hair follicle stem cells and used two different types of experiments to track their division. The experiment found that while young mice had normal and healthy symmetrical and asymmetric divisions, an unusual type of cell division appeared in older mice.
To further investigate the reasons for this, the research team looked at the role of a group of proteins called hemidosomes that attach cells to the extracellular matrix and help them understand their position. The team’s analysis showed that in older mice, these proteins become increasingly unstable and produce a different and abnormal cell type during the division of hair follicle stem cells.
The result was that hair follicle stem cells were destroyed with age, leading to their removal from the body. As a result, over time, this can lead to thinning and loss of hair follicles.
According to the study’s lead author, Hiroyuki Matsumura, these dramatic results show how hair follicles lose their ability to regenerate over time.
